Offline Enrichment of Primary Aromatic Amines with SPE technique and investigation of their migration into artificial saliva solution with LC-MS/MS technique

Hidden risks related to manmade products often get to be discovered with the growing knowledge in our civilised world. For instance, many additives are applied for different purposes during the production of food packaging materials. Among the colorants, the group of azodyes is a potential source of danger as they can release primary aromatic amines under reductive conditions. Regulation No. 2011/10/EC contains a list of chemicals that are allowed to use in the production of food contact materials and also demands to carry out migration studies into food simulants in order to determine the migration level of the listed compounds....The maximum level of migration in the case of primary aromatic amines is 10 μg/kg expressed as aniline equivalent. Directive No. 2002/61/EC is also related to primary aromatic amines and restricts the use of 22 compounds in leather and textile products that can come into contact with human skin or the oral cavity for a prolonged time. This regulation draws special attention to risks relating to small children but no further precaution has been enacted.

Small children are likely to take everything into their mouths that is at their hands. Thus, it might be crucial to investigate the migration of the above mentioned compounds from different kinds of colorful plastic toys into an artificial saliva solution which composition is similar to the saliva of infants. As the European Union determined an extremely low migration limit our research requires an analytical method that has high sensitivity such as LC-MS/MS. However, further enrichment is inevitable, hence the aim of my work is to develop an offline and rugged SPE technique using isotope labeled internal standards suitable for the determination and enrichment of 24 primary aromatic amines from artificial saliva solution. I intend to study the migration of the target compounds from colorful plastic products right after the successfull validation of the method.
